不同强度跑台运动通过调控 BDNF/miR-124/p-CREB信号通路改善 CUMS小鼠抑郁样行为

摘要:

[目的]基于慢性应激诱导的抑郁动物模型,探讨不同强度连续跑台运动对缓解小鼠抑郁行为的干预作用及可能机制。[方法]8周龄雄性健康KM小鼠,随机分为空白对照组(SED)、慢性不可预知应激刺激(CUMS)抑郁模型组(SMD)、抑郁+低强度运动组(LICT) 、抑郁+高强度运动组(HICT),每组15只。空白对照组和模型组常规喂养,不进行运动;模型运动组进行8周不同强度跑台运动干预。运动干预结束后测定各组小鼠的神经行为评分,实时荧光定量RT-PCR检测miR-124及相关mRNA的表达水平,Western blot检测海马BDNF、p-CREB以及RGS4的蛋白表达水平。[结果]LICT和HICT组均有效改善了CUMS实验小鼠的抑郁和焦虑样行为。与对照组相比,运动组miR-124和RGS4呈现差异负相关表达,且差异表达量与不同强度运动相关。不同强度的运动可有效改善抑郁小鼠海马功能,上调BDNF、p-CREB及RGS4的表达水平,下调miR-124的表达水平。[结论]低强度和高强度运动通过影响BDNF/CREB信号通路改善CUMS小鼠抑郁样行为,低强度连续运动可能是缓解焦虑和抑郁的有效方式。

[Obiective] To explore the effects and possible mechanisms of different intensities of continyous treadmill running on chronic unpredictable mild stress(CUMS) depression mouse model. [Methods] Healthy 8-week-old male KM mice were randomly divided into the normal(SED), the model( SMD), the low-intensity exercise group(LICT), and high-intensity exercise group(HICT), 15 mice in each group. The normal group and the model group were fed regularly and did not exercise, the exercise group were placed on the treadmill for 8 weeks with different exercise intensity. After the exercise intervention, the neurobehavioral scores of each group of mice were measured; the expression levels of miR-124 and related mRNA were detected by RT-PCR; the protein expression levels of BDNF, p-CREB and RGS4 in hippocampus were detected by Western blot.[Results] Both the LICT and HlCT groups efectively improved the depression and anxiety-like behaviors of CUMS mice. Compared with the control group, the exercise group showed a negative correlation between miR-124 and RGS4 expression, and the differential expression levels were associated with different intensities of exercise. Different intensities of exercise can effectively improve hippocampal function in CUMS mice ,upregulating the expression levels of BDNF, p-CREB, and RGS4, and downregulating the expression level of miR-124. [Conclusion] Low-intensity and high-intensity exercise can improve depressive-like be havior in CUMS mice by affecting the BDNF/CREB signaling pathway, low-intensity continuous exercise may be an effective way to alleviate anxiety and depression.
